Overview

Description

SOLIScript® 1-step CoV Kit is optimized for highly sensitive detection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) viral RNA by one-step real-time RT-PCR (RT-qPCR).

The Kit is suitable for the detection of up to four targets simultaneously and can be used with all common qPCR cyclers. Follow recommendations specific for a selected SARS-CoV-2 diagnostic panel, and instrument-specific guidelines. Contact us if a passive reference dye is required for signal normalization.

SARS-CoV-2 primers and probes, applicable positive and internal extraction controls are not provided with the Kit and should be supplied by the user. If full solution is needed, please check out: SOLIScript® SARS-CoV-2 RT-qPCR Multiplex Assay Kit.

SOLIScript® 1-step CoV Kit is tested and validated for SARS-CoV-2 detection by diagnostic laboratories. Please see our recommendations for US-CDC and Berlin-Charité protocols. 

Note! Amplification of more than one target in a single reaction may require considerable optimization, individual assays must be validated prior to multiplexing. For multiplex reactions, design primer pairs with similar annealing temperatures for all targets to be amplified. Self-complementarity of primers should be avoided.

Does RiboGrip help with inhibitor resistance?

RiboGrip™ is not directly associated with tolerance to common inhibitors, however, it helps to keep RNA intact and prevent RNA degradation because RiboGrip is inactivating ribonuclease enzymes

Which primes, probes are recommended for COVID-19 detection with Your 1-step kit?

The Kit has been tested and validated by diagnostic laboratories using primer-probe panels recommended by Charité, (Germany) and CDC (USA) protocols according to WHO “Coronavirus disease (COVID-19) technical guidance: Laboratory testing for 2019-nCoV in humans”, section: “Molecular assays to diagnose COVID-19”; subsection: “In-house developed molecular assays”

Are primers, probes and positive control included in the 1-step RT-qPCR CoV kit?

SARS-CoV-2 primers and probes, applicable positive and internal extraction controls are not provided with the Kit and should be supplied by the user.

Why do certain kits contain a ROX?

ROX (carboxy-X-rhodamine) is one of the passive reference dyes providing a constant fluorescent signal that is used for signal normalization during the amplification cycles. The emission recorded from a reference dye during the baseline cycles is used to normalize the emission recorded from the reporter (e.g. EvaGreen®, SolisGreen®, SYBR® Green, etc.) in later cycles in ROX-dependent real-time PCR systems (e.g. Applied Biosystems 5700, 7000, 7300, 7700, 7900HT, StepOne™, StepOnePlus™). Reference dye compensates for small fluorescent fluctuations and well-to-well variations that may occur.

In Solis BioDyne qPCR mixes we use technology, based on ROX, that allows us to use same mix for high- and low- ROX requiering cyclers.
